GLP Facilities

The 16,000 ft2 CBB GLP laboratory/office offers state-of-the-art fermentation/bioprocessing
equipment and an experienced professional full-time staff with expertise
in process development. For example, our contract culturing capabilities
include:
 |
Large-capacity multi-tier shakers,
in environmentally controlled rooms, |
 |
Eight two-liter fermentors, used
for optimization, |
 |
A total of 23 fermentors to 1,000-liter
working capacity, |
 |
A liquid-oxygen supply system for
high-cell-density culture at 100- and 1,000-liter scale, and |
 |
A tandem 25-ton glycol
cooling system. |

As an example of capabilities, CBB has performed
100-liter fermentations with in-situ microfiltration, as well as
fermentations with simultaneous product removal by adsorption.
CBB has performed many unique chromatographic separations to recover
valuable products. State-of-the-art downstream capabilities match
the scale of the fermentors. A summary of holdings is outlined
below, and tabulated details follow the summary.
 |
Fundamentals include a large medium
preparation room, sterilizers, and washer. Multi-tier, high-capacity
shakers are housed in 29°C and 37°C incubation rooms.
The laboratory also contains two 4°C cold rooms, as well
as supporting infrastructure, including an explosion-proof solvent-extraction
room with walk-in hood, laboratory benches, etc. |
 |
Principal fermentors are listed
in Table 1. Most fermentors have PC-based, on-line measurement
and control capabilities. All have systems capable of handling
explosive and flammable gases as well as aseptic sample collection
and inoculation. Most fermentors have on-line, off-gas analysis
capability. In addition, a clean-in-place system is available
to service large fermentors. A liquid-oxygen system is available
to provide high rates to 100-and 1,000-liter high-cell-density
fermentations. In addition, a high-capacity glycol cooling system
can rapidly chill fermentors and holding tanks. |
 |
Analytical services are partially
listed in Table 2. The liquid chromatography unit is both analytical
and preparative scale and has processed up to 36 L/h. |
 |
CBB is fully prepared
for downstream, pilot-scale operations in fermentation, harvesting,
and bioprocessing. For example, the filtration system has recovered
low-molecular weight materials while processing more than 200
L/h. Table 3 provides a partial listing of downstream processing
capabilities. |
 |
CBB has several protein purification systems. For purifications under GLP status, CBB has an Akta Explorer Chromatography system and an Akta Pilot chromatography system. . . . more |
