Answer to Practice Problem #2 Noncompartmental Pharmacokinetics Tutorial:
The first step is to calculate the MTT for each route of administration, using the equation MTT = AUMC/AUC:
|
Form of Administration |
AUC (mg-hr/L) |
AUMC (mg-hr2/L) |
MTT (hr) |
|
Intravenous bolus |
52 |
290 |
5.6 |
|
Oral tablet, Manufacturer 1 |
52 |
345 |
6.6 |
|
Oral tablet, Manufacturer 2 |
45 |
297 |
6.6 |
Recall that after an i.v. bolus dose, MTT = MRT. For this drug, the MRT = 5.6 hr. Moreover, after an oral dose,
MTT = MRT + MAT
Rearranging:
MAT
= MTT - MRT
Thus, for Manufacturer 1: MAT = MTT – MRT = 6.6 hr – 5.6 hr = 1.0 hr
And, for Manufacturer 2: MAT = MTT – MRT = 6.6 hr – 5.6 hr = 1.0 hr
Obviously, these MAT values are identical.
